In vitro expression of ncRNAs and RNA-binding proteins and localization of RNP complexes in vivo

I am involved in the functional investigation of macromolecular associations between proteins and long non-coding RNAs (ncRNAs). In particular, my project is focused on the expression and purification of brain-specific long ncRNA targets and RNA-binding proteins. For this purpose T7 RNA polymerase-based in vitro transcription and cell-free in vitro translation methods will be used for RNA and protein, respectively. In addition, fluorescence techniques (FISH, FRET) will be applied to investigate possible co-localisation and tissue-specific expression of RNA and protein targets in vivo.
